SS-31, also known as Elamipretide, is a synthetic tetrapeptide with the sequence D-Arg-2’6′-Dmt-Lys-Phe-NH2. It belongs to a class of cell-permeable, mitochondria-targeted peptides originally developed to study mitochondrial membrane dynamics. SS-31 is characterized by alternating aromatic and cationic residues that allow it to concentrate within the inner mitochondrial membrane.
Research has examined SS-31 for its potential to interact with cardiolipin – a phospholipid unique to the inner mitochondrial membrane that is essential for optimal electron transport chain function. In laboratory settings, studies have explored how SS-31 may help stabilize cardiolipin structure, thereby supporting mitochondrial cristae integrity and ATP production efficiency.
Preclinical research has examined SS-31 in models of cardiac ischemia-reperfusion injury, heart failure, and age-related mitochondrial dysfunction. Studies have also explored its effects in renal injury models and neurodegenerative disease contexts where mitochondrial dysfunction plays a prominent role. Researchers have investigated its potential to reduce reactive oxygen species (ROS) production without acting as a traditional antioxidant.
