Humanin is a 21-amino acid peptide encoded within the 16S ribosomal RNA region of the mitochondrial genome – a discovery that expanded the understood protein-coding capacity of the human mitochondrial genome. It is secreted and functions as an autocrine, paracrine, and endocrine signaling molecule, acting through a receptor complex involving CNTFR, WSX-1, and gp130 – components shared with cytokine signaling pathways.

Researchers have studied Humanin for its cytoprotective and anti-apoptotic properties, particularly in neuronal cells. In vitro studies have examined its ability to inhibit amyloid-beta (A?)-induced neuronal apoptosis, making it a compound of interest in Alzheimer’s disease biology research.
