GHRP-6

GHRP-6 (Growth Hormone Releasing Peptide-6) is a synthetic hexapeptide with the sequence His-D-Trp-Ala-Trp-D-Phe-Lys-NH2. It was one of the first synthetic growth hormone secretagogues developed, acting as a functional ghrelin mimetic and agonist at the GHSR-1a receptor. Its development was pivotal in understanding non-GHRH pathways for GH regulation.

Researchers have studied GHRP-6 extensively for its ability to stimulate GH release through the ghrelin receptor, which is distinct from the GHRH receptor pathway. Research has examined the additive effects of combined GHRP-6 and GHRH administration on pulsatile GH secretion in animal models. Studies have also explored how GHRP-6 may augment somatotroph sensitivity to endogenous GHRH.

A notable characteristic researchers have examined is GHRP-6’s pronounced appetite-stimulating effects, which appear to be mediated through central ghrelin receptor activation in hypothalamic feeding centers. This orexigenic property has made it useful in research models studying the ghrelin/appetite axis. Researchers have also explored GHRP-6’s potential gastroprotective effects in preclinical models of gastric mucosal injury, consistent with ghrelin receptor biology in the gastrointestinal tract.
