Sermorelin is a synthetic 29-amino acid peptide corresponding to the biologically active N-terminal fragment of endogenous growth hormone-releasing hormone (GHRH). The full native GHRH molecule consists of 44 amino acids, but research has demonstrated that the first 29 residues retain the full capacity to bind and activate GHRH receptors on pituitary somatotroph cells.

Researchers have studied Sermorelin for its ability to stimulate the pituitary gland’s natural, pulsatile secretion of growth hormone. Unlike exogenous GH administration, Sermorelin works upstream by stimulating the pituitary’s own production mechanisms, which remain subject to normal negative feedback regulation via somatostatin. Research has examined how this mechanism preserves the physiological pulsatility of GH release.

Preclinical studies have explored Sermorelin’s effects on GH and IGF-1 levels in animal aging models where endogenous GHRH signaling may be attenuated. Research has also examined its potential interactions with hypothalamic GHRH neurons and feedback regulation of the GH axis. Its shorter half-life compared to GHRH analogues like CJC-1295 has made it a useful tool for studying acute pituitary responsiveness to GHRH stimulation.
