CJC-1295 + Ipamorelin Blend is a research formulation combining a modified GHRH analogue (CJC-1295) with a selective growth hormone secretagogue (Ipamorelin). These two compounds work through entirely distinct receptor systems – GHRH receptors and ghrelin/GHSR-1a receptors respectively – to stimulate GH release through complementary pathways. Research has characterized their co-administration as producing synergistic amplification of GH pulse amplitude.

Researchers have studied the mechanistic basis of CJC-1295 and Ipamorelin synergy. CJC-1295 (No DAC form) stimulates GHRH receptors on pituitary somatotrophs to enhance their sensitivity and readiness to secrete GH, while Ipamorelin activates GHSR-1a to directly trigger GH release. Studies have compared the GH output following combined administration to either compound alone, examining whether the effects are truly synergistic at the pituitary level or simply additive.

Preclinical research using this combination has examined GH peak amplitude, IGF-1 levels, and pulse frequency in animal models. Studies have explored how the combined stimulation of both the GHRH and ghrelin receptor pathways affects the total amount of GH released and the duration of the GH response. Research has also examined whether Ipamorelin’s selectivity advantage – minimal effects on cortisol and prolactin – is preserved when combined with CJC-1295, making this blend a useful research tool for studying pituitary GH secretion biology.
