5-Amino-1MQ (5-Amino-1-methylquinolinium) is a small molecule research compound that acts as a potent, selective inhibitor of nicotinamide N-methyltransferase (NNMT). NNMT is an enzyme expressed primarily in adipose tissue and liver that methylates nicotinamide (a form of vitamin B3), consuming S-adenosylmethionine (SAM) in the process. Its inhibition has been explored as a strategy to influence cellular metabolic pathways.

Researchers have studied 5-Amino-1MQ for its potential to increase NAD+ precursor availability by reducing NNMT-mediated nicotinamide consumption. Research has explored how NNMT inhibition might influence the methionine cycle and SAM availability, potentially affecting epigenetic methylation patterns and one-carbon metabolism. Studies have examined whether reduced NNMT activity correlates with changes in adipocyte differentiation and lipid accumulation in cell culture models.

Preclinical research in mouse models has examined 5-Amino-1MQ’s effects on body composition, adipose tissue metabolism, and energy expenditure. Studies have explored its influence on mitochondrial function in fat cells and skeletal muscle, as well as its effects on NAD+-dependent enzyme activity including sirtuins. Research has also examined its potential interactions with mTOR signaling pathways relevant to cellular metabolism and growth.
