Hexarelin is a synthetic hexapeptide with the sequence His-D-2-MeTrp-Ala-Trp-D-Phe-Lys-NH2. It belongs to the growth hormone secretagogue (GHS) peptide class and acts as a potent, selective agonist at the growth hormone secretagogue receptor (GHSR-1a), commonly known as the ghrelin receptor. It was among the early synthetic GHS peptides developed for research into GH axis regulation.
Research has examined Hexarelin for its potent stimulation of GH release from pituitary somatotrophs through GHSR-1a activation. Studies have compared its GH-releasing potency to other secretagogues and examined the degree of GH release it can elicit in various animal models. Researchers have explored whether Hexarelin-induced GH release is additive or synergistic with GHRH co-administration in research settings.
Beyond GH secretion, researchers have examined Hexarelin’s direct cardiac effects through GHSR-1a receptors expressed in cardiac tissue. Preclinical studies have explored its potential cardioprotective properties in models of cardiac ischemia and heart failure. Research has also examined its influence on cortisol and prolactin secretion, distinguishing it from more selective GH secretagogues. Its high potency has made it a valuable pharmacological tool for studying GHSR-1a receptor biology.
